O foco central do laboratório de RPN é a exploração das ligações entre metabolismo > transcrição > splicing alternativo > células estaminais/ doença, alicerçado nos princípios fundamentais da bioquímica e dando ênfase a translação e aplicação clínica na área das terapias celulares. Recentemente o laboratório demonstrou que a modulação bioquímica e física da estrutura da cromatina pode constituir uma ferramenta poderosa para alterar o processo de transcrição e expressão génica. Modulando um simples parâmetro bioquímico, a pressão osmótica extracelular, é possível alterar a estrutura da cromatina e a transcrição. Um estímulo Hypo-osmótico contribui para o relaxamento da cromatina e induz alterações na actividade da RNA polymerase II, constituindo uma opção para manipular a tomada de decisão em termos de estado celular. Esta tecnologia foi testada em protocolos de indução da pluripotência e de trans-diferenciação, em células que crescem em suspensão e/ ou agarradas a substratos, CD34+ MNCs do sangue do cordão umbilical, fibroblastos e células B, com acréscimo de eficiência. Alem disso, quando as células são cultivadas em substratos de baixa rigidez exibem um incremento de zonas de cromatina aberta e expressão de genes que facilitam a indução de pluripotência. O grupo é especialista na síntese e entrega de nanopartículas com diferentes cargas em células primárias e estaminais e patenteou uma NP ativável pela luz, capaz de libertar ácido retinóico dentro do nicho leucémico e induzir a diferenciação de células iniciadoras de leucemia. Neste contexto o grupo está a explorar a libertação de intermediários metabólicos capazes de modular nichos estaminais e microambientes tumorais no sentido de desenvolver novas terapias celulares para o cancro e medicina regenerativa.
