As barreiras corporais permitem que os organismos interajam com o mundo circundante para sentir, respirar, reproduzir e atender às suas necessidades nutricionais. No entanto, falhas nessas barreiras podem levar a doenças infecciosas e transformação neoplásica.
No intestino, uma interação saudável entre as células imunes, células epiteliais, dieta e micróbios é fundamental para preservar a integridade da barreira epitelial intestinal, ao mesmo tempo que forma uma linha de frente de defesa contra patógenos invasores.

As células T convencionais requerem cerca de uma semana após o encontro com o antigénio para fornecer proteção eficaz. Em contraste, as células T inatas adquirem propriedades efetoras pré-programadas durante o desenvolvimento no timo, sendo preparadas para uma resposta rápida e imediata às mudanças no epitélio. No entanto, pouco se sabe acerca de como este programa é iniciado.

O grupo explora como as células T inatas percebem as sinais de dieta e como estes são integrados no seu desenvolvimento e função. No passado recente, a equipa mostrou que, além de seu programa genético regulado pelo desenvolvimento, as células do sistema imunológico são controladas por estímulos ambientais (Nature 2014; Cell Reports 2015 e J Immunol 2016).

A equipa concentra-se principalmente em desvendar os mecanismos pelos quais os micronutrientes controlam o desenvolvimento de células T inatas e função nas mucosas, como o intestino, explorando as vias moleculares intrínsecas subjacentes em situação de saúde e doença.

Abordamos essas questões combinando tecnologias celulares e moleculares multifacetadas in vivo e in vitro. Isso inclui genética de ratinho de última geração, citometria de fluxo, microscopia confocal, assim como perfis de expressão genética de subpopulações imunológicas e epiteliais.
